I am a GS employee. Been employee for 13 years.

My wife has a self-employed business. I help her out but not in her payroll/employee status.

We're thinking about making her business into S-Corp. Can I be on partnership or employee with her business as a GS employee? Is there a law prohibit Fed Employees to get a 2nd job or start a business?

You can, but there are ethic rules you need to follow.

You can’t work that on govt time
You can’t use govt equipment to do that job nor solicit your side business.
You need to stay away from influencing things like being involved in contracting nor being in a situation where your job crosses paths with that company of your wife’s sch as in business decisions.
You will likely not be able to adjust your duty time to fit this other job.
If this company has a clear political bent that might show your political leanings it could hurt your career when as a fed you are to be non partisan
In lobbying this company you can not use your fed govt position in any form of influence

It would be a terrible mistake if you didn't get guidance/clearance in writing from your agency's Ethics official before you do this.
